بررسی اثر سمیت سلولی اسانس گیاه آویشن وحشی با استفاده از روش های MTT و تریپان بلو بر رده سلولی سرطان سینه MCF-7

سرطان سینه شایع ترین سرطان در بین زنان ایرانی است. اسانس آویشن وحشی دارای ترکیبات ضد سرطان همچون تیمول می باشند. هدف از این تحقیق مطالعه اثر اسانس این گیاه بر رده سلول سرطانی سینه MCF-7 توسط روشهای MTT و تریپان بلو و مقایسه آن با تاکسول به عنوان یک ترکیب شناخته شده ضد سرطان می باشد. رده سلولی سرطان سینه MCF-7 و و رده طبیعی فیبروبلاست HU-02 (خریداری شده از مرکز ملی ذخائر ژنتیکی و زیستی ایران) با غلظت های مختلف اسانس آویشن وحشی و تاکسول (به عنوان شاهد مثبت) در فواصل زمان های 24، 48 و 72 ساعت معین تیمار شدند. میزان سمیت سلولی با روش MTT و تریپان بلو بدست آمد. نتایج نشان داد اسانس آویشن وحشی مورد مطالعه دارای اثر سیتوتوکسیک بصورت وابسته به دوز در رده سلولی MCF-7 است. بیشترین اثر سایتوتوکسیک اسانس این گیاه در غلظت 125 میکروگرم بر میلی لیتر بر سلولهای سرطان سینه بود در حالیکه در هیچ غلظت مورد مطالعه بر سلولهای غیر سرطانی اثر سوء نشان نداد. از طرف دیگر بررسی نشان داد اسانس آویشن وحشی می تواند در کاربرد توام با تاکسول٬ سمیت سلولی تاکسول بر سلولهای سرطانی را تشدید کند، بنابراین ممکن است در درمان سرطان سینه موثر باشد و احتمالا از طریق القاء آپوپتوز باعث مرگ سلول می شود.
