Association of Melatonin Receptor 1A Gene Polymorphisms with Production and Reproduction Traits in Zandi Sheep

Melatonin regulates some major physiological processes such as maturation and function of reproductive system, pubertal development, seasonal reproduction and adaptation. The activation of melatonin hormone is mediated by melatonin receptor. Previous studies showed melatonin receptor 1A gene (MTNR1A) is highly polymorphic in seasonally breeding species. The aim of the present study was detection of MTNR1Apolymorphism and its association with production (body weights at birth, 1, 3, 6, 9 and 12 month of age) and reproduction (litter size) traits in Zandi sheep using PCR-RFLP methodology. Hundred blood samples were randomly collected and genomic DNA was isolated using modified salting out method. A large fragment of exon 2 of MTNR1A gene was amplified by PCR using specific primer pairs. After amplification, the PCR product was digested with MnII endonuclease. Restricted digestion allowed the determination of two alleles (M, m) and two genotypes (MM, Mm) with frequencies of 0.91, 0.09 and 0.82, 0.18, respectively. Least square means showed that MM individuals had higher body weight at one month of age (BW1) than Mm individuals (P<0.05). No associations were found between observed genotypes and other studied traits.In the subsequent studies using a large number of samples along with the other important parameters such as sire effect and the lamb health status which may influence lamb body weight is recommended.
ملاتونین برخی فرآیندهای اصلی فیزیولوژیک از قبیل بلوغ و عملکرد سیستم تولیدمثلی، رشد و توسعه بلوغ و تولیدمثل فصلی و سازگاری را کنترل میکند. فعالسازی هورمون ملاتونین به واسطه گیرنده ملاتونین انجام میشود. مطالعات پیشین نشان دادهاند که ژن گیرنده شماره یک-A ملاتونین (MTNR1A) در نژادهای دارای تولیدمثل فصلی بسیار چند شکل میباشد. هدف مطالعه حاضر ردیابی چند شکلیهای MTNR1A و ارتباط آنها با صفات تولیدی (اوزان بدن در تولد، 1، 3، 6، 9 و 12 ماهگی) و تولیدمثلی (تعداد بره) در گوسفند زندی با استفاده از روش PCR-RFLP بوده است. صد نمونه خون به طور تصادفی جمعآوری شده و DNA با روش نمکی بهینه یافته استخراج شد. یک قطعه بزرگ از اگزون شماره 2 ژن MTNR1A به وسیله PCR و جفت آغازگرهای اختصاصی تکثیر شد. بعد از تکثیر، محصولات PCR با اندونوکلئاز MnlI هضم شدند. هضم محدود شده امکان شناسایی دو آلل (M و m) و دو ژنوتیپ (MM و Mm) را به ترتیب با فراوانیهای 91/0، 09/0 و 82/0، 18/0 فراهم آورد. میانگین حداقل تفاوتها نشان داد که افراد MM دارای وزن بدن بیشتری در یک ماهگی (BW1) نسبت به افراد Mm (05/0>P) بودند. هیچ ارتباطی بین ژنوتیپهای مشاهده شده و سایر صفات مشاهده نشد. در مطالعات بعدی برای جایگاه نشانگری MTNR1A تعداد بزرگتری از نمونهها در کنار سایر پارامترهای مهم از قبیل اثرات نرها و وضعیت سلامت برهها که احتمالاً وزن بدن برهها را تحت تأثیر قرار میدهند پیشنهاد میشوند.
