اثر شش هفته تمرین هوازی و عصاره دارچین بر بیان ژن لپتین در بافت کبد موش های نر تغذیه شده با رژیم غذایی پرچرب

چکیده مقاله :
دو عامل سطح لپتین سرم به واسطه تاثیر بر اشتها و توانایی کاهش انرژی دریافتی و میزان فعالیت بدنی به واسطه توانایی افزایش انرژی مصرفی، نقش بسیار مهم و تعیین کنندهای در کنترل وزن و توده چربی بدن دارند. مصرف برخی ترکیبات گیاهی ضد چاقی نظیر دارچین نیز می تواند در کاهش توده چربی بدن و کنترل چاقی موثر و مفید باشد. پژوهش تجریی حاضر با هدف تعیین اثرات اجرای شش هفته تمرین هوازی (پنج روز در هفته)، مصرف عصاره دارچین (200 میلی گرم به ازای هر گیلوگرم از وزن بدن) و غذای پرچرب (150 میلی لیتر امولسیون غذای پرچرب مازاد بر رژیم غذایی روزانه) بر بیان ژن لپتین در بافت کبد موش های نر تغذیه شده با رژیم غذایی پرچرب در قالب یک طرح پژوهشی چند گروهی با گروه کنترل انجام شد. برای بررسی میزان بیان ژن یا mRNA پروتئینهای مورد نظر از روش Real Time-PCR استفاده شد. اثرات رژیم غذایی پرچرب, ترکیب رژیم غذایی پرچرب و تمرین هوازی، ترکیب رژیم غذایی پرچرب و دارچین و ترکیب رژیم غذایی پرچرب و تمرین هوازی و دارچین بر متغیر لپتین در بافت کبد چشمگیر و معنی دار بود, به طوری که مصرف غذای پرچرب باعث تغییرات نامطلوب بیان ژن شده ولی اجرای شش هفته تمرین و مصرف عصاره دارچین باعث تعدیل اثرات نامطلوب رژیم غذایی پرچرب شده و تغییرات مطلوبی را در بیان ژن و وزن بدن و وزن بافت کبد ایجاد کرده اند. به علاوه ترکیب اجرای شش هفته تمرین و عصاره دارچین به مراتب اثرات مطلوب تری را در تعدیل اثرات نامطلوب رژیم غذایی بر بیان این ژن داشته است. بنابراین توصیه می شود که همراه با اجرای تمرینات ورزشی هوازی از عصاره دارچین نیز استفاده شود.
چکیده انگلیسی:
Given their effect on appetite and the ability to reduce energy intake and physical activity, serum leptin levels play a key and decisive role in controlling body weight and fat mass. Consumption of some anti-obesity plant compounds such as cinnamon can also be useful in reducing body fat mass and controlling obesity. This experimental study was aimed at determining the effects of six weeks of aerobic exercise (five days a week), consumption of cinnamon extract (200 mg per kg of body weight) and high-fat food (150 ml of high-fat food emulsion in excess of daily diet). Expression of leptin gene in the liver tissue of male rats fed a high-fat diet was performed in a multi-group research design with a control group. Real Time-PCR was used to evaluate the expression of the gene or mRNA of the proteins. Takara Viragen’s kit was employed to prepare PCR master mix for cDNA. The effects of high-fat diet, combination of high-fat diet and aerobic exercise, combination of high-fat diet and cinnamon and combination of high-fat diet, aerobic exercise, and cinnamon on leptin variable in liver tissue were considerable and significant so that high-fat diet caused adverse changes in the expression of this gene. Nevertheless, six weeks of exercise and consumption of cinnamon extract moderated the adverse effects of a high-fat diet and caused favorable changes in the expression of this gene and body weight and liver tissue weight. In addition, the combination of six weeks of exercise and cinnamon extract had far more favorable effects in modulating the adverse effects of diet on the expression of this gene. Hence, it is recommended to use cinnamon extract along with aerobic exercise. .
