بررسی تاثیر باکتری اشرشیا کُلی (Escherichia coli) بر بافت بیضه در موش صحرایی

بیماریهای عفونی دستگاه تناسلی شامل میکروبها و ویروسهایی است که از طریق ارتباط جنسی منتقل میشوند. سفلیس و سوزاک عفونتهای قدیمی تناسلی هستند، اما امروزه بیماریهای دیگری مانند ایدز و آنتیژنهایی که از طریق رابطه جنسی منتقل میشوند نیز مطرح هستند. هدف از مطالعه حاضر بررسی تاثیر باکتریاشرشیا کلی بر بافت بیضه و لوله های اسپرم ساز و پارامترهای مربوط به اسپرم بود. تعداد 16 سر موش صحرایی نر بالغ نژاد ویستار با وزن متوسط 200 تا 220 گرم با سن 5/2 تا 3 ماه به دو گروه 8 تایی تقسیم شده و در طی 60 روز دوره مطالعه، حیوانات در معرض 12ساعت تاریکی و 12ساعت روشنایی قرار گرفتند. سوش اشرشیاکلی انتروتوکسیژنیک (سروتایپ-0114) از آزمایشگاه مرجع بوعلی تهران تهیه و جهت عفونی نمودن موشها استفاده شد. پس از ایجاد عفونت از بافت اسپرم نمونه آسیب شناسی تهیه و با رنگ آمیزی هماتوکسیلین ـ ائوزین بررسی شد. همچنینن میزان تحرک، قابلیت زیست و تعداد اسپرمها نیز ارزیابی گردید. نتایج مطالعه بافتی نشان داد در گروه شاهد تمامی لوله های اسپرم ساز به همدیگر چسبیده و تمامی رده های سلولهای زاینده جنسی قابل مشاهده بود. درحالیکه در گروه دریافت کننده اشرشیا کلی، لوله های اسپرم ساز از هم گسیخته شده و رده های سلولی جنسی از بین رفته اند. نتایج بررسی پارامترهای اسپرم نشان داد، که میزان تعداد اسپرم، قابلیت زیست اسپرم و تحرک اسپرم بصورت معنی داری در گروه آلوده به باکتری ای کلی نسبت به گروه شاهد کاهش می یابد. نتایج مطالعه نشان داد که باکتری اشرشیا کلی بصورت مشخصی بر میزان باروری و بافت بیضه تاثیر نامطلوب داشته و میتواند باعث کاهش باروری گردد.
There is more kind of reproductive infectious diseases. Viruses and bacterial agents can cause to presents this disease that occurs by sexual contact. Syphilis and Gonorrhea are the most common old known diseases that affects urogenital tract system, in nowadays HIV and transmitted Antigens by sexual contacts are the others reasons of urinary infectious diseases. The aim of current study was to investigate the effects of E. coli on testis tissue, seminiferous tubules and semen parameters. Materials and methods: sixteen mature male Wistar rats with 200-220 grams’ weight and 2.5- 3 months’ ages, distributed to two of 8 rat’s groups and for 60 days of study period the lightning program was 12 hours’ light and 12 hours’ dark. Entro-toxigenic E coli (0114 serotype[n1] [PB2] ) was gained from Tehran Bouali reference laboratory for infect rats. After infection, samples were taken from testis tissue and Hematoxylin-eosin was used for histopathology. The motility, viability and semen count was evaluated. Results: Histopathology studies indicated that in control group all seminiferous tubules were adhere together and all sexual germinal cells was seen. In E. coli infected group, seminiferous tubules were inconsecutive and germinal cells was not seen. Semen parameters evaluation showed that total count of sperm, viability and motility was decreased significantly in E. coli infected group in comparison to control group. The results indicated that the E. coli has undesirable effects on fertility rate and testis tissue and could decrease fertility rate.
