خالص سازی و خصوصیات سرولوژیکی فیتوپلاسمای برگ سفیدی نیشکردر استان خوزستان

چکیده مقاله :
بیماری برگ سفیدی نیشکر یکی از مخربترین بیماریهای نیشکر در آسیا میباشد. این بیماری به عنوان یک بیماری فیتوپلاسمایی بر روی نیشکر در استان خوزستان نیز گزارش شده است. علاوه بر نیشکر، مرغ نیز علائم برگ سفیدی ناشی از فیتوپلاسما را در این استان نشان میدهد. به منظور خالصسازی نسبی فیتوپلاسمای عامل برگ سفیدی از نیشکر آلوده به بیماری استفاده شد. آموده حاصل از خالصسازی نسبی 40 گرم بافت آلوده به خرگوش تزریق و بعد از خونگیری، آنتی سرم تهیه گردید. آنتی سرم حاصل پس از جذب با عصاره سالم در آزمایشهای الیزا (PTA-ELISA)، دیبا (DIBA) و (TPIA) Tissue Print، فقط قادر به تشخیص و شناسایی فیتوپلاسمای همولوگ خود بود درحالیکه هیچ گونه واکنشی بین آنتی سرم با فیتوپلاسماهای غیر همولوگ (زردی پروانش، جاروک لیمو ترش، جاروی بادام و برگ سفیدی مرغ) مشاهده نشد.
Sugarcane white leaf (SCWL) is among the most important and economical diseases of sugarcane in Asia. SCWL is reported from Khuzestan province as an emerging phytoplasmal disease of sugarcane. In addition to SCWL, Bermuda grass white leaf (BGWL) the other monocot phytoplasmal disease also is reported from this province. Rabbit has been used for rising of antiserum by injection of partially purified SCWL at 40g of infected tissue. This antiserum exhibited specificity for its homologous phytoplasma antigen in Plate- Trapped Antigen enzyme-linked immunosorbent assay (PTA-ELISA) dot immunoblotting assay (DIBA) and tissue print immuno assay (TPIA). No cross-reactions were observed in reciprocal tests between this antiserum and other tested phytoplasmas.
