اثر پرایمینگ بذر با سدیم اسکوربات بر جوانه زنی و برخی ویژگی های فیزیولوژیکی کنجد (Sesamum indicum) در دماهای مختلف

به منظور بررسی تاثیر آسکوربات و دما بر میزان آنزیم های آنتی اکسیدان و شاخص های جوانه زنی کنجد آزمایشی در سال 1393 در آزمایشگاه تحقیقاتی دانشکده علوم کشاورزی دانشگاه محقق اردبیلی به صورت فاکتوریل در قالب طرح کاملاً تصادفی در سه تکرار انجام گرفت. تیــمارهای آزمایشی شـامل سه سطح آسکوربات (شاهد، 5 و 10 میلی مولار) و دماهای مختلف جوانه زنی (15، 25 و 35 درجه سلسیوس) بودند. صفـات اندازه گیری عبارت از شاخص های جوانه زنی، میزان آنزیم های آنتی اکسیدان، پرولین و پروتئین بودند. نتایج نشان داد که کاربرد آسکوربات منجر به افزایش پرولین و کاهش میزان آنزیم ها گردید، و در دمای 15 درجه سلسیوس نسبت به دماهای 25 و 35 درجه سلسیوس میزان آنزیم ها افزایش یافت. همچنین، کاربرد آسکوربات موجب کاهش تخریب پروتئین ها گردید. از آن جایی که کنجد گیاهی گرمادوست است به نظر می رسد افزایش میزان آنزیم های آنتی اکسیدان در دمای 15 درجه سلسیوس به دلیل افزایش تولیـد رادیکال های آزاد در این دما می باشد. همبستگی مثبت و معنی دار آنــزیم های آنتی اکسیدان رابطه مستقیم این صفات را نشان می دهد. همچنین همبستگی مثبت و معنی داری بین شاخص های جوانه زنی مشاهده شد. با توجه به نتایج حاصل از این پژوهش به نظر می رسد کاربرد آسکوربات آسیب های ناشی از دماهای نامطلوب را کاهش می دهد و تاثیر مثبتی نیز بر شاخص های جوانه زنی دارد. همچنین، خاصیت آنتی اکسیدانی آسکوربات نیاز به فعالیت آنزیم های آنتی اکسیدان را کاهش داده و به رشد گیاهچه های کنجد کمک می کند.
To study the effects of ascorbate and temperature on antioxidant enzymes and germination indexes, a factorial experimental based on completely randomized block design with three replications was conducted at the Agricultural Research Laboratory of Mohaghegh Ardabili University in 2014. Treatments were three levels of ascorbate (control, 5 and 10 mM) and three levels of temperatures (15, 25 and 35 °C). Parameters under study were germination indices, production rates of antioxidant enzymes, proline and protein. The results of experiment indicated that ascorbate application that of raised proline content. Application of ascorbate resulted in enzymes reduction at 15°C more than 25°and 35° temperatures. Its application also reduced protein denaturation. Since sesame is a warm season plant, it seems that increasing antioxidant enzymes at 15°C is due to higher production of free radicals. The positive and significant correlation between antioxidant enzymes and germination indices represents their direct relationships. According to the results of this experiment, it seems that the application of ascorbate had positive effect on germination indices and reduced the effects of improper temperatures; it also benefited seedling growth of sesame.
