1 - بررسی مقایسهای آناتومی برگ ودمبرگ در سه گونه گلابی در استان گیلان (Pyrus L.)فصلنامه بیولوژی کاربردی , العدد 4 , السنة 10 , پاییز 1399هدف:هدف این پژوهش بررسی مقایسهای صفات آناتومیکی برگها و دمبرگهای 3 گونه گلابی استان گیلان (تلکا، گرگانیPyrus boissieriana Buhse ، خج Pyrus communis L. و خزری Pyrus hyrcana Fedor ) میباشد. مواد و روشها:در این پژوهش برای مطالعه آناتومیکی گونهها، ابتدا نمونههای أکثرهدف:هدف این پژوهش بررسی مقایسهای صفات آناتومیکی برگها و دمبرگهای 3 گونه گلابی استان گیلان (تلکا، گرگانیPyrus boissieriana Buhse ، خج Pyrus communis L. و خزری Pyrus hyrcana Fedor ) میباشد. مواد و روشها:در این پژوهش برای مطالعه آناتومیکی گونهها، ابتدا نمونههای گیاهی جمعآوری شدند. سپس روش برشگیری دستی در آزمایشگاه انجام شد. برای رنگآمیزی نمونهها رنگآمیزی کارمن مورد استفاده قرارگرفت. پس از رنگآمیزی، از سطح مقطع برشها لام تهیه شد و برای مشاهده مقاطع عرضی برگها و دمبرگها از میکروسکوپ نوری مجهز به عدسی مدرج استفاده گردید. سپس اطلاعات حاصل از صفات آناتومی آنالیز آماری شدند. یافتهها:ضخامت برگ و دمبرگ در بین 3 گونه مورد بررسی متفاوت بود. بیشترین ضخامت رگبرگ میانی در بین سه گونه گلابی مطالعه شده مربوط به گونه p . communis (11/1±682 میکرومتر) بود و کمترین ضخامت رگبرگ میانی، در p . hyrcana (05/1±66/431 میکرومتر) مشاهده شد. صفات آناتومیکی در تفکیک گونه، مؤثر و دارای اهمیت میباشند. بنابراین، خصوصیات آناتومیکی در گونههای مختلف گیاهی از نظر ردهبندی تاکسونومیکی مفید خواهد بود. تفاصيل المقالة -

This study established an effective protocol for callus induction and bulblet regeneration in A. stamineum using various explant types (radicle, basal plate, and cotyledon) and growth regulators. The results indicated that the best callus formation from cotyledon explants occurred in media containing 1 mg/L 2,4-D, 1 mg/L 2,4-D with 0.5 mg/L BA, 2 mg/L 2,4-D with 0.5 mg/L kinetin, and 1 or 2 mg/L 2,4-D with 1 mg/L kinetin. In terms of regeneration, cotyledon explants showed the highest regeneration rate compared to radicle and basal plate explants, with 3.33 regenerations per explant in MS medium supplemented with 1 mg/L kinetin and 1 mg/L NAA. Additionally, the highest bulblet regeneration rate (11 per explant) was obtained from callus on PGR-free medium. The maximum number of roots (7.90) and root length (10.9 mm) were observed on MS medium containing 3 mg/L IBA. Rooted bulblets were successfully acclimatized in pots filled with a cockpit mixture (3:1 v/v) with a 100% survival rate. This study not only provides a successful in vitro propagation technique for A. stamineum but also facilitates its breeding program. تفاصيل المقالة
