زمینه و هدف: آنزیم کربنیک انهیدراز متالوآنزیم حاوی یون روی (Zn+2) است که هیدراسیون برگشت پذیر دی اکسید کربن را به یونهای بی کربنات و هیدروژن کاتالیز میکند. عملکرد این آنزیم در انجام بسیاری از فرآیندهای فیزیولوژیک بدن مانند تنفس، تعادل اسید-باز، ترشح الکترولیتها، کلسی فیکاسیون، پیام رسانی سیستم عصبی و بیوسنتز لیپیدها نقش مهمی ایفا میکند. در این مطالعه نقش مهاری سدیم اگزالات و اتیلن دی آمین بر آنزیم کربنیک انهیدراز انسانی و الگوی مهاری آن بررسی شد. روش بررسی: چهار غلظت متفاوت از اتیلن دی آمین و سدیم اگزالات: 20، 100، 500 و 1000 میکرومولار انتخاب گردید و فعالیت آنزیم در عدم حضور و حضور این ترکیبات بررسی گردید. به منظور تعیین دقیق پارامترهای سنیتیک کربنیک انهیدراز معادله جفت معکوس لاینویور-برگ استفاده گردید. همچنین میزان IC50 مهارکنندهها بدست آمد. نتایج: نمودارهای جفت معکوس لاینویور-برگ نشان داد دو ترکیب فوق نقش مهاری بر فعالیت آنزیم کربنیک انهیدراز دارند و نوع مهار برای اتیلن دی آمین و سدیم اگزالات به ترتیب به صورت رقابتی و ضد رقابتی است. مقادیر Km، Vmax، IC50 و Ki نشان داد سدیم اگزالات اثر مهاری قویتری نسبت به اتیلن دی آمین دارد. نتیجهگیری: براساس نتایج بدست آمده اتیلن دی آمین و سدیم اگزالات موجب مهار آنزیم کربنیک انهیدراز میشوند. همچنین سدیم اگزالات اثر مهارکنندگی قویتر نسبت به اتیلن دی آمین دارد. بنابراین باید در میزان مصرف آنها بخصوص در صنعت دقت لازم به عمل آید. زیرا اختلال در عملکرد آنزیم کربنیک انهیدراز منجر به اختلال در عملکرد فرآیندهای فیزیولوژیک و بروز برخی بیماریها گردد.

آلفاگلوکزیداز از گروه آنزیمهای هیدرولازی است که انتهای غیراحیایی کربوهیدراتها را به آلفاگلوکز هیدرولیز میکند. مهار این آنزیم موجب مهار جذب گلوکز میگردد. ترکیباتی که آنزیم آلفاگلوکزیداز را مهار میکنند جهت درمان دیابت نوع دو به کار میروند. گیاه خارخسک (Tribulus terrestris) گیاه دارویی است که در سراسر جهان رویش دارد. ساپونینهای این گیاه ترکیب مؤثر اصلی آن و مسئول فعالیتهای بیولوژیکی گیاه میباشند. در این پژوهش تاثیر غلظتهای مختلف ساپونین استخراج شده از گیاه خارخاسک بر فعالیت و ساختار آلفاکلوگوزیداز توسط طیفسنجی الایزاریدر، UV-Visible و دورنگ نمایی دورانی (CD) مورد بررسی قرار گرفت. نتایج طیفسنجی الایزاریدر نتایج نشان داد ساپونین به صورت ضد رقابتی موجب مهار فعالیت آنزیم آلفاگلوکزیداز میگردد. مقدار نصف غلظت مهاری (IC50) برای ساپونین 38 میکرومولار محاسبه شد. جذب نور مرئی-فرابنفش آلفاگلوکزیداز با افزودن غلظتهای ساپونین افزایش یافت. مقایسه طیف CD آنزیم آلفاگلوکزیداز و کمپلکس ساپونین-آنزیم نشان داد محتوای ساختار آلفا هلیکس کاهش و محتوای ساختارهای صفحات بتا و رندوم کویل افزایش یافت. در مجموع، نتایج نشان داد ساپونین استخراج شده از گیاه خارخسک تغییرات ساختاری در آلفاگلوکوزیداز ایجاد میکند و فعالیت آنزیم را کاهش میدهد. به صورت خلاصه، ساپونینهای استخراج شده از گیاه خارخاسک میتواند به عنوان کاندیدایی مناسب جهت طراحی داروی درمان دیابت نوع دوم باشد.

Purpose: The objective of this investigation was to evaluate the synergistic effect of paclitaxel (PTX) combined with titanium dioxide nanoparticles (TiO2NPs) on DNA structure and to examine the proliferation of MDA-MB-231cells.Methods: This investigation performed with Ultraviolet spectroscopy, zeta potential investigation, circular dichroism (CD) spectroscopy, ELISA reader and fluorescence spectroscopy. Results: The Ultraviolet results indicated that the structure of DNA in the presence of PTX and TiO2NPs (at a lower concentration) changed significantly rather than TiO2NPs or PTX alone. The fluorescence results exposed that PTX+TiO2NPs could form a complex via non-intercalative mechanism and the PTX+TiO2NPs affinity to DNA increased considerably. The thermodynamics parameters displayed that PTX+TiO2NPs interact with DNA strongly and in this interaction, the hydrophobic force plays an important role. The CD data confirmed that DNA structure was modified by PTX+TiO2NPs via a simple and reasonable mechanism: change in DNA conformation from B to C-form. The negative charge of DNA reduced strongly after addition of PTX+TiO2NPs. The anticancer property of PTX+TiO2NPs by MTT assay demonstrates that this combination can tremendously diminish the proliferation of MDA-MB-231cells compared to PTX or TiO2NPs alone.Conclusion: Based on this investigation TiO2NPs could enhance the affinity and binding of PTX (at a lower concentration) on DNA structure and PTX+NDs can promote mortality of MDA-MB-231 cells. This study can offer an innovative strategy for designing the ideal anti-tumor agents.

Background & Aim:Saponins are well-known secondary metabolites with numerous beneficial pharmacological properties. Since the interactions of drugs with blood constituents, in particular with human serum albumin (HSA) may have a major impact on drug pharmacology, the present study designed to provide a fundamental understanding of the interaction of saponins extracted from Tribulus terrestris (typically utilized in folk medicine) with HSA in detail. Experimental: After extraction and purification of saponins; UV-Vis spectroscopy, fluorescence spectroscopy, and far-UV CD spectroscopy were used to examine the effects of saponins from Tribulus terrestris on HSA. Results: Thin-Layer chromatography confirmed the presence of 3 different saponins. The UV-Vis absorption, fluorescence emission and far-UV CD results displayed that saponinsfrom Tribulus terrestris could form a complex with HSA. The binding constant for the saponins–HSA complex was found to be 13.4×104M-1. The distance r between HSA and Tribulus terrestris saponins was also acquired according to the Förster theory. Recommended applications/industries: Altogether, saponins can bind to HSA and change the secondary and tertiary structure of HSA moderately. The results obtained from this study can help in understanding the pharmacokinetic properties of saponins.
